Background
PRKCB2 is a calcium-activated, phospholipid and diacylglycerol (DAG)-dependent serine/threonine protein kinase involved in various cellular processes, such as regulation of the B cell receptor (BCR) signaling complex, oxidative stress-induced apoptosis, androgen receptor-dependent transcriptional regulation, insulin signaling, and endothelial cell proliferation. It plays a crucial role in B cell activation by regulating BCR-induced NF-κB activation. PRKCB2 mediates activation of the canonical NF-κB pathway (NFKB1) by directly phosphorylating Ser-559, Ser-644, and Ser-652 sites of CARD11/CARMA1. Phosphorylation induces CARD11/CARMA1 to bind to lipid rafts and recruits the BCL10-MALT1 complex and MAP3K7/TAK1, thereby activating the IKK complex, leading to NFKB1 nuclear translocation and activation. Furthermore, PRKCB2 also plays a direct role in the negative feedback regulation of BCR signaling by directly phosphorylating BTK at the Ser-180 site. BTK phosphorylation alters BTK plasma membrane localization, thereby inhibiting BTK activity.
