Background
ROS1 is a receptor tyrosine kinase (RTK) involved in epithelial cell differentiation and regionalization of the proximal epididymal epithelium. NELL2 serves as an endogenous ligand for ROS1. Upon stimulation by NELL2, ROS1 activates intracellular signaling pathways, triggering epididymal epithelial differentiation and subsequent sperm maturation (By similarity). It may activate several downstream signaling pathways related to cell differentiation, proliferation, growth, and survival, including the PI3 kinase-mTOR pathway. ROS1 mediates the phosphorylation of PTPN11, an activator of this pathway. Additionally, it may phosphorylate and activate the transcription factor STAT3 to regulate anchorage-independent cell growth. ROS1 also mediates the phosphorylation and activation of VAV3, a guanine nucleotide exchange factor involved in cell morphology regulation. Other downstream signaling proteins potentially activated by ROS1 include AKT1, MAPK1, MAPK3, IRS1, and PLCG2.
Verified Bioactivity
The activity was measured by off-chip mobility shift assay(MSA). The enzyme was incubated with fluorecence-labeled substrate and Mg(or Mn)/ATP. The phosphorylated and unphosphorylated substrates were separated and detected by MSA device. When the protein concentration is 3.125 nM, the conversion rate is about 23%.
