Syndecan-3 Protein, Human (HEK293, His) is a Dendritic cells (DCs)-specific heparan sulfate (HS) proteoglycan that captures HIV-1 through interaction with the HIV-1 envelope glycoprotein gp120. Description

Syndecan-3 Protein, Human (HEK293, His) is a Dendritic cells (DCs)-specific heparan sulfate (HS) proteoglycan that captures HIV-1 through interaction with the HIV-1 envelope glycoprotein gp120. SDC3 is involved in the regulation of growth factor signalling, Notch and BMP signalling, adhesion and migration, proliferation and differentiation. Syndecan-3 is associated with the control of feeding behaviour and the generation of cerebellar fibrillar plaques in Alzheimer鈥檚 disease[1][2][3]. Syndecan-3 Protein, Human (HEK293, His) is the recombinant human-derived Syndecan-3 protein, expressed by HEK293 , with C-6*His labeled tag.

Background

Syndecans (sdcs) are heparan sulphate proteoglycans (HSPG) composed of a core protein to which heparan sulphate (HS) glycosaminoglycan chains are covalently attached. These molecules form part of the glycocalyx, which comprises a network of membrane-bound proteoglycans and glycoproteins at the cell surface of endothelial cells. There are four mammalian syndecans, designated syndecan-1 (sdc-1), -2, -3, and -4. Sdc-3 is the predominant syndecan in the nervous system, where it was first identified, and has been associated with the control of feeding behaviour and the generation of cerebellar fibrillar plaques in Alzheimer’s disease. Sdc-3 is also an HSPG of the musculoskeletal system[1].
Syndecan-3 stabilizes the captured virus, enhances Dendritic cells (DCs) infection in cis, and promotes transmission to T cells. Removal of the HSs from the cell surface by heparinase III or by silencing syndecan-3 by siRNA partially inhibited HIV-1 transmission by immature DCs, whereas neutralizing both syndecan-3 and DC-SIGN completely abrogated HIV-1 capture and subsequent transmission. Syndecan-3 (SDC3) has the largest core protein of all four mammalian syndecans and can harbour both heparan sulphate and chondroitin sulphate chains[2][3].

Verified Bioactivity

Measured by the ability of the immobilized protein to inhibit the adhesion of Saos‑2 human osteosarcoma cells to human Fibronectin. The ED50 for this effect is 0.5-2.0 μg/ml in the presence of 5 μg/mL Fibronectin.
