Background

Tissue plasminogen activator (t-PA) is a pivotal protein that catalyzes the conversion of the abundant, yet inactive, zymogen plasminogen into the active protease plasmin, achieved by hydrolyzing a single Arg-Val bond in plasminogen. This enzymatic process is integral in controlling plasmin-mediated proteolysis, thereby playing a crucial role in tissue remodeling, degradation, and cell migration, as well as influencing various physiopathological events. Furthermore, t-PA exhibits functional significance during oocyte activation, participating in the cortical granule reaction in the zona reaction, which contributes to the prevention of polyspermy and serves as a protective mechanism during fertilization (By similarity). The multifaceted roles of t-PA underscore its importance in both physiological and reproductive processes, emphasizing its regulatory functions in proteolysis and cellular events.

Biological Activity

Measured by its ability to cleave a peptide substrate, N-carbobenzyloxy-Gly-Gly-Arg-7-amido-4-methylcoumarin (Z-GGR-AMC). Read at excitation and emission wavelengths of 380 nm and 460 nm (top read). The specific activity is >500.47 pmol/min/µg, as measured under the described conditions.
