SYK-1106 dihydrochloride has not only the highest efficacy (Emax = 111%) and potency (EC50 = 0.089 nM) for the DOR but also almost no agonist activities toward the MOR and KOR[1].
SYK-1106 dihydrochloride binds to DOR, KOR and MOR, with Ki values of 0.848 nM, 2.45 nM, and 9.54 nM, respectively[1].

In Vivo

SYK-1106 (0.3 mg/kg, s.c.) dihydrochloride exhibits potent, dose-dependent antidepressant-like effects without convulsions effects in the mouse forced-swimming tests[1].

Animal Model: Male ICR mice (4-6 weeks old) bearing forced swim test[1]
Dosage: 0.3 mg/kg
Administration: s.c.
Result: Showed dose-dependent antidepressant-like effects in the mouse forced-swimming test, reducing immobility and increasing swimming and climbing behaviors.
No convulsive effects were observed at the effective dose.
