1. Preparation of stock solution and working solution
Prepare a high-concentration stock solution to dilute into the working buffer.
Dissolve BCDA in a suitable solvent, such as DMF or DMSO. The typical concentration of the stock solution is 10-50 mg/mL.
Buffer: Prepare a suitable buffer for the esterase (for example, 50 mM Tris-HCl, pH 7.5).
Dilution: Dilute the stock solution into the buffer, with the final concentration being 0.1-1 mg/mL (or approximately 0.5-5 mM).
2. Detection method
2.1 Incubation: Incubate the sample at 37°C or room temperature.
2.2 Visualization: The esterase activity will convert the substrate into a blue precipitate.
2.3 Monitoring: Monitor the development of color. The reaction time depends on the enzyme activity, usually ranging from 30 minutes to several hours.
