2-PMPA prodrug 1 (Compound 18b) (10 mg/kg; p.o.; single administration) effectively overcomes the extremely poor oral absorption of 2-PMPA, and generates µM levels of 2-PMPA in mouse plasma. However, it cannot efficiently deliver 2-PMPA into the CNS, which limits its application in neurological disorders but makes it suitable for peripheral GCPII targets^[1].
