    Flow cytometric analysis of 1X10^6 MCF-7 cells labeling CXCR7 Antibody(red). Cells were fixed with 4% paraformaldehyde and permeabilised with 0.2% Triton X-100. Then stained with the primary antibody at 1/400 dilution overnight at 4℃.Alexa Fluor® 488 Goat Anti-mouse IgG H&L (invitrogen A11001) was used as the secondary antibody at 1/1,000 dilution for 45 minutes at room temperature. Mouse IgG Isotype Control (Invitrogen 14-4714-B2, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (gray).

Alexa Fluor® 488 Goat Anti-mouse IgG H&L (invitrogen A11001, green) was used as the secondary antibody at 1/500 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. The Nuclear counterstain was DAPI (Blue)

    GPCR RDC1/CXCR-7 is an Atypical chemokine receptor that controls chemokine levels and localization via high-affinity chemokine binding that is uncoupled from classic ligand-driven signal transduction cascades, resulting instead in chemokine sequestration, degradation, or transcytosis. Also known as interceptor (internalizing receptor) or chemokine-scavenging receptor or chemokine decoy receptor. Acts as a receptor for chemokines CXCL11 and CXCL12/SDF1. Chemokine binding does not activate G-protein-mediated signal transduction but instead induces beta-arrestin recruitment, leading to ligand internalization and activation of MAPK signaling pathway. Required for regulation of CXCR4 protein levels in migrating interneurons, thereby adapting their chemokine responsiveness. In glioma cells, transduces signals via MEK/ERK pathway, mediating resistance to apoptosis. Promotes cell growth and survival. Not involved in cell migration, adhesion or proliferation of normal hematopoietic progenitors but activated by CXCL11 in malignant hemapoietic cells, leading to phosphorylation of ERK1/2 (MAPK3/MAPK1) and enhanced cell adhesion and migration. Plays a regulatory role in CXCR4-mediated activation of cell surface integrins by CXCL12. Required for heart valve development. Regulates axon guidance in the oculomotor system through the regulation of CXCL12 levels; (Microbial infection) Acts as a coreceptor with CXCR4 for a restricted number of HIV isolates[1][2][3][4][5][6][7][8][9][10][11][12][13].

    Tissue_specificity:It is expressed in monocytes, basophils, B cells, umbilical vein endothelial cells (HUVECs) , and B lymphoblasts. It is expressed at lower levels in CD4+ T lymphocytes and natural killer cells. In brain tissue, it can be detected in endothelial cells and capillaries, as well as in mature neurons of the frontal cortex and hippocampus. It is expressed during renal tubule formation in the kidneys. It is highly expressed in astrocytoma endothelial cells, microglia, and glioma cells. It is expressed at low levels in normal CD34+ progenitor cells but at high levels in several myeloid malignant cell lines. It is expressed in breast cancer but not in normal breast tissue (protein level) .
