Sections were blocked with 5% goat serum and incubated overnight at 4 °C with the following primary antibodies: anti-α-smooth muscle actin (α-SMA) (HY-P80484, 1:200), anti-F4/80 (F4/80 Antibody (YA002), HY-P80665, 1:200), anti-Integrin α5 (ITGA5) (HY-P80416, 1:200), and anti-Desmocollin-2 (DSC2) (1:200). Representative immunofluorescence staining images of iNOS and Arg-1 in RAW 264.7 cells and the quantification analysis of iNOS and Arg-1 expression. To investigate whether NVP-BHG712 affects macrophage polarization, macrophages were labeled with anti-F4/80 (F4/80 Antibody (YA002), overnight at 4 °C) antibody, while M1 and M2 macrophages were labeled with anti-iNOS and anti-Arg-1 antibodies, respectively. Immunofluorescence staining results showed that LPS stimulation enhanced polarization toward the iNOS+ M1 phenotype and significantly inhibited polarization toward the Arg-1+ M2 phenotype.

Co-immunofluorescence staining of CLEC4E (red) and F4/80 (green) in colonic tissues (scale bar: 100 µm). 5 μm-thick frozen colonic tissue slides and pre-fixed cell samples were incubated with 5% BSA in PBS at room temperature for 1.5 h to block nonspecific binding. Then the above samples were exposed to primary antibodies specific for CLEC4E (1:200), F4/80 Antibody (YA002) (1:100) overnight in a 4°C refrigerator. After that, the specimens were washed three times in PBS and incubated with fluorescently labeled secondary antibodies for 1.5 h at room temperature under dark conditions. Following further PBS rinses, nuclear staining was performed with DAPI, and confocal fluorescence images were collected using a Leica microscope.

Immunocytochemistry analysis of RAW 264.7 cells labeling F4/80 with F4/80 Antibody (HY-P80665) at 1/100 dilution. Cells were fixed in 4% paraformaldehyde for 15 minutes at room temperature, permeabilized with 0.1% Triton X-100 in PBS for 15 minutes at room temperature, then blocked with quick block buffer for 10 minutes at room temperature. Cells were then incubated with F4/80 Antibody (HY-P80665) at 1/100 dilution in quick block buffer overnight at 4 ℃. AF488-conjugated Goat Anti-Rat IgG H&L (HY-P89199, Green) was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. The Nuclear counterstain was DAPI (Blue). -
Flow cytometric analysis of 1X106 RAW264.7 cells labeling F4/80 Antibody (HY-P80665, red). Cells were stained with the primary antibody at 1/50 dilution for an hour at 4℃. AF488-conjugated Goat Anti-Rat IgG H&L (HY-P89199) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Rat IgG2a Isotype Control (HY-P89510, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black).
