Arachidonoyl-L-carnitine chloride
Arachidonoyl-L-carnitine (C20:4 Carnitine) chloride is an arachidonic acid-L-carnitine complex and also a substrate for β-oxidation. Arachidonoyl-L-carnitine chloride undergoes mitochondrial β-oxidation, a process that requires energy-dependent function provided by NADP+ transhydrogenase supplying NADPH for 2,4-enoyl-CoA reductase activity. Arachidonoyl-L-carnitine chloride shows reduced respiration under uncoupling conditions, and ATP partially restores its oxidation.

Arachidonoyl-L-carnitine chloride (40 μM) β-oxidation by isolated control rat liver mitochondria is inhibited by uncoupling conditions, with respiration rates reduced by ~36% in malonate-containing medium and ~40% in malate-containing medium compared to ADP-coupled rates[1].
Arachidonoyl-L-carnitine chloride (40 μM) β-oxidation by isolated clofibrate-treated (0.2% w/w diet for 12 days) rat liver mitochondria is no longer inhibited by uncoupling conditions, with respiration rates similar to or slightly higher than ADP-coupled rates in both malonate- and malate-containing media[1].
Arachidonoyl-L-carnitine chloride (40 μM) uncoupler-inhibited β-oxidation by isolated control rat liver mitochondria is potently rescued by ATP, increasing respiration rates to 9 ng-atoms of O/min per mg of protein, which is more than double the rate achieved with ADP[1].
