Firefly luciferase-IN-1 is a cell-permeable firefly luciferase inhibitor with an IC50 of 0.25 nM and a Ki of 1.72 nM. Firefly luciferase-IN-1 reversibly binds to the D-luciferin-occupied pocket of Fluc, competing with D-luciferin. Firefly luciferase-IN-1 effectively blocks Fluc/D-Luciferin (HY-12591A)-induced bioluminescence.

In Vitro
Firefly luciferase-IN-1 (compound 48) is a highly potent inhibitor of purified Firefly luciferase, with an IC50 of 0.25 nM[1].
Firefly luciferase-IN-1 (20 μM) selectively inhibits Fluc without affecting Rluc activity, and its inhibitory effect is not due to absorption of luminescence[1].
Firefly luciferase-IN-1 (1.0 μM) reversibly inhibits Firefly luciferase by competitively binding to the D-luciferin pocket, with a KI of 1.72 nM[1].
Firefly luciferase-IN-1 inhibits Fluc in transiently transfected 293T cells with an IC50 of 2.6 nM and an IC50 of 15 nM in cell lysates[1].
Firefly luciferase-IN-1 (compound 48) potently inhibits intracellular Fluc in PC9-Fluc, A549-Fluc, H1299-Fluc, Hepa1-6-Fluc, and BEAS2B-Fluc cell lines, with IC50 values ranging from 2.1 to 32 nM[1].
