Folitixorin calcium (5,10-Methylenetetrafolate calcium; ANX-510) is a reduced form of Folic acid (HY-16637) and a cofactor of thymidylate synthase. Folitixorin calcium acts as an essential cofactor to promote the formation of a tight ternary complex between thymidylate synthase and FdUMP, thereby enhancing the inhibitory effect of FdUMP on thymidylate synthase. Folitixorin calcium in combination with 5-fluorouracil and αVEGF reduces tumor volume in colorectal tumor models of nude mice. Folitixorin calcium is used in related research on Yoshida sarcoma, P-388 leukemia, and colorectal cancer.

Folitixorin (1000 ns) calcium forms structurally conserved stable complexes with human thymidylate synthase and Klebsiella pneumoniae thymidylate synthase ^[2].

In Vivo

Folitixorin (0.6 mg/mouse; i.p.; once daily for five consecutive days) calcium in combination with 5‑Fluorouracil and αVEGF reduces tumor volume and improves survival rate in a colorectal tumor model of nude mice^[3].

Animal Model:

6-8 week old nude mice (nu/nu) with 2 x 10⁶ HT-29 cells^[3]
Dosage:

i.p., daily, five consecutive days with the exception of αVEGF, dosed once (100 μg/mouse) on day 1
Result:

Reduced tumor volume and improved survival rate in a colorectal tumor model of nude mice.
