Description

Linoleic acid is a critical component of polyunsaturated fatty acids. Linoleic acid reduces GSH-PX and T-SOD activities. Linoleic acid functions as a structural component to maintain a certain level of membrane fluidity of the transdermal water barrier of the epidermis. Linoleic acid can cause acute oxidative damage to red blood cells, ultimately leading to partial acute anemia. Linoleic acid can be used in the research of skin diseases associated with linoleic acid deficiency[1][2].

In Vivo

Linoleic acid (0.5 mL, 15-90 mmol/L; i.v.; single tail vein injection) induces dose-dependent acute oxidative red blood cell injury in male Sprague-Dawley rats, with the 60 mmol/L dose causing the most significant reductions in Hb (16.8 g/L) and RBC (0.71×1012/L) at 24 h post-injection, along with suppressed antioxidant enzyme activity and increased ferryl Hb formation[1].

Animal Model: Sprague-Dawley (male, 200±20 g)[1]
Dosage: 0.5 mL, 15 mmol/L; 0.5 mL, 30 mmol/L; 0.5 mL, 60 mmol/L; 0.5 mL, 90 mmol/L
Administration: i.v.; single tail vein injection
Result: Reduced Hb by 13.5±2.0 g/L and RBC by 0.45±0.15×1012/L at 24 h post-injection (30 mmol/L dose).
Decreased GSH-PX, T-SOD, and H2O2 levels significantly compared to controls (P < 0.05) at 24 h and 48 h post-injection (30 mmol/L dose).
Induced red blood cell morphological changes (spindle shapes, pleomorphism, shrinkage, deformation, rupture), most distinct at 24 h post-injection (30 mmol/L dose).
Reduced Hb by 16.8±1.5 g/L and RBC by 0.71±0.18×1012/L at 24 h post-injection (60 mmol/L dose).
Decreased GSH-PX, T-SOD, and H2O2 levels significantly compared to controls (P < 0.05 at 24 h, P < 0.01 at 48 h) post-injection (60 mmol/L dose).
Detected a significant absorbance peak at 425 nm (ferryl Hb Soret peak) at 24 h post-injection, with peak intensity declining at 48 h and 72 h (60 mmol/L dose).
Caused 50% of rats to die within 5 minutes of injection, presenting with dyspnea, cyanosis, labial haemoptysis, and convulsions (90 mmol/L dose).
Reduced Hb and RBC levels at 24 h post-injection, with changes less severe than 30 mmol/L and 60 mmol/L doses (15 mmol/L dose).
