Administration: Murine cardiac TnI peptide (250 μg, emulsified in complete Freund's adjuvant (CFA) (HY-153808)), subcutaneous (s.c.) immunization on day 0 and day 7 • anti‑mouse PD‑1 antibody (5‑10 mg/kg), intraperitoneal (i.p.) injection every 2 days for total 5 doses starting from day 7/8
(2) Anti-PD-1 dose and start day differ slightly between protocols (5 mg/kg from day 7[3]; 10 mg/kg from day 8[2]); dose titration recommended.
Serum myocardial injury markers: Elevated CK, elevated CK‑MB, elevated LDH‑1, elevated cTnI, elevated cTnT
Histology analysis: Myocardial inflammatory infiltration, cardiomyocyte necrosis, interstitial fibrosis; detected by H&E, Masson's trichrome, Sirius Red staining
Molecular changes: Elevated NLRP3, cleaved caspase‑1, cleaved GSDMD, IL‑1β, IL‑18 in heart tissue; activated NF‑κB pathway with elevated p‑p65, elevated p‑IκBα, elevated p65 DNA‑binding activity; reduced HIF‑1 mRNA
Survival: Significantly increased mortality compared with vehicle group
