Patent Blue V calcium salt is a triarylmethane dye used for tissue staining and lymphatic tracing, mainly applied by topical injection or eye drops. Patent Blue V calcium salt has affinity for specific tissues (such as corneal endothelium, lymphatic system), and stains the target structure by adsorption or binding, assisting in precise operation during surgery. Patent Blue V calcium salt is mainly used in ophthalmic surgery (such as graft staining for Descemet's membrane endothelial keratoplasty) and lymphatic drainage localization for sentinel lymph node biopsy of tumors.

Biological Activity
Guide (Following is our recommended protocol. This protocol only provides a guideline, and should be modified according to your specific needs).
Patent Blue V (2.5%; 10 minutes) can maintain staining for more than 25 minutes in Descemet membrane endothelial keratoplasty (DMEK) graft staining experiments. Patent Blue V has a slightly lower staining intensity than Trypan blue (HY-D0970) and no significant difference in endothelial cell loss rate, making it a potential alternative dye to Trypan blue[1].
Patent Blue V staining results can be detected by UV-visible spectrophotometry or fluorescence spectroscopy, and its maximum absorption wavelength is about 630 nm. The specific experimental steps are as follows[2]:
1. Sample preparation: Dissolve Patent Blue V calcium salt in physiological saline or balanced salt solution (BSS) to prepare stock solution, and use a serial dilution method (such as 1/2 gradient) to prepare a concentration gradient solution.
2. Instrument calibration: Use a UV-visible spectrophotometer (such as Unicam UV/visible spectrophotometer), calibrate the baseline with a blank solvent (such as BSS), set the scan range to 326-700 nm, and the scan interval to 1 nm.
3. Spectral scanning: Take 100 μL of each concentration solution and inject it into the quartz cuvette, scan the absorption spectrum in the range of 326-700 nm, and record the absorbance (A) value. The typical absorption peak appears near 630 nm, confirming the maximum absorption wavelength.
4. Data verification: Compare the absorption curves of solutions of different concentrations to verify that the peak at 630 nm increases with increasing concentration, ensuring that there is no interference from other impurities.
