  3. PD-1/PD-L1-IN-65

PD-1/PD-L1-IN-65 is a PD-1/PD-L1 checkpoint inhibitor, with an EC50 of 6.06 μM for blocking the PD-1/PD-L1 interaction and a Kd of 1.43 μM for PD-L1. PD-1/PD-L1-IN-65 upregulates pro-inflammatory cytokines IFN-γ, IL-1α, IL-1β and the cytolytic protein perforin, and reverses T cell exhaustion. PD-1/PD-L1-IN-65 enhances T cell-mediated cancer cell lysis. PD-1/PD-L1-IN-65 can be used in the research of breast cancer.

In Vitro

PD-1/PD-L1-IN-65 (Compound 17h) functionally blocks the interaction between PD-1 and PD-L1 in competitive ELISA assays, with an EC50 value of 6.06 μM[1].
PD-1/PD-L1-IN-65 (8 μM; 0.0327-8 μM) effectively restores T cell signaling in cell-based PD-1/PD-L1 blockade assays, with an EC50 of 215 nM[1].
PD-1/PD-L1-IN-65 (10 μM) reverses PD-L1-mediated immunosuppression in the MDA-MB-231/activated PBMC co-culture system by upregulating IFN-γ, IL-1α, IL-1β, and perforin[1].
PD-1/PD-L1-IN-65 (5-10 μM) enhances T cell-mediated lysis of MDA-MB-231 cells without intrinsic cytotoxicity, confirming its selective immunomodulatory activity[1].

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
