Description

The BACE1 protein is an important member of the aspartic protease family and is critical in Alzheimer's disease pathology.Alternative splicing generates multiple transcript variants, at least one of which encodes a preproprotein that is proteolytically processed to produce the mature protease.BACE1 Protein, Human (HEK293) is the recombinant human-derived BACE1 protein, expressed by HEK293 , with tag free.

Background

BACE1, a member of the peptidase A1 family of aspartic proteases, plays a pivotal role in Alzheimer's disease pathology. Through alternative splicing, this gene generates multiple transcript variants, with at least one encoding a preproprotein that undergoes proteolytic processing to yield the mature protease. As a transmembrane protease, BACE1 catalyzes the initial step in the production of amyloid beta peptides from the amyloid precursor protein. Amyloid beta peptides constitute the primary component of the amyloid beta plaques that accumulate in the brains of individuals with Alzheimer's disease. The expression of BACE1 is widespread, showing ubiquity across various tissues, including the pancreas, brain, and 24 other tissues, underscoring its significance in diverse physiological contexts.

Verified Bioactivity

Measured by its ability to cleave a fluorogenic peptide substrate, Mca-SEVNLDAEFRK(Dpn)RR-NH2. Read at excitation and emission wavelengths of 320 nm and 405 nm. The specific activity is >1.5 pmol/min/µg, as measured under the described conditions.

Product Properties

Appearance

Lyophilized powder

Formulation

Lyophilized from a 0.22 μm filtered solution of 50 mM Tris-HCl, 100 mM NaCl, pH 8.0.
Note: For SPR assay, please replace the buffer. Primary amine components (e.g., Tris, imidazole) can affect protein-coupled chips.

Endotoxin Level

<1 EU/μg, determined by LAL method.

Reconstitution

It is not recommended to reconstitute to a concentration less than 100 μg/mL in ddH2O.

Storage & Stability

Stored at -20°C for 2 years from date of receipt. After reconstitution, it is stable at 4°C for 1 week or -20°C for longer (with carrier protein). It is recommended to freeze aliquots at -20°C or -80°C for extended storage.
