Background

BACE1, a member of the peptidase A1 family of aspartic proteases, plays a pivotal role in Alzheimer's disease pathology. Through alternative splicing, this gene generates multiple transcript variants, with at least one encoding a preproprotein that undergoes proteolytic processing to yield the mature protease. As a transmembrane protease, BACE1 catalyzes the initial step in the production of amyloid beta peptides from the amyloid precursor protein. Amyloid beta peptides constitute the primary component of the amyloid beta plaques that accumulate in the brains of individuals with Alzheimer's disease. The expression of BACE1 is widespread, showing ubiquity across various tissues, including the pancreas, brain, and 24 other tissues, underscoring its significance in diverse physiological contexts.

Biological Activity

Measured by its ability to cleave a fluorogenic peptide substrate, Mca-SEVNLDAEFRK(Dpn)RR-NH2. Read at excitation and emission wavelengths of 320 nm and 405 nm. The specific activity is 1.79 pmol/min/µg, as measured under the described conditions.
