Cat. No.: HY-177646A
Synonyms: ALN-APP sodium
Mivelsiran (ALN-APP) sodium is an siRNA targeting the β-amyloid precursor protein (APP). Mivelsiran sodium induces sequence-specific degradation of APP mRNA via RNA interference, thereby reducing the levels of APP protein and downstream amyloid cleavage products. Mivelsiran sodium reduces β-amyloid protein levels in the brain, alleviates neuroinflammation, and attenuates microglial proliferation. Mivelsiran sodium can be used for research on early-onset Alzheimer's disease .

Cas9 Nickase D10A mRNA expresses a version of the Streptococcus pyogenes SF370 Cas9 protein (CRISPR Associated Protein 9) that contains a D10A amino acid substitution. This mRNA also contains a C-terminal nuclear localization signal followed by a HA tag.Cas9 functions as part of the CRISPR (clustered regularly interspaced short palindromic repeats) genome editing system. In the CRISPR system, an RNA guide sequence targets the site of interest and the Cas9 protein is employed to perform the DNA cleavage. While wild-type Cas9 creates a double-stranded break at the target site, Cas9 nickase creates a single-stranded break. This favors homology-directed repair and decreases the occurrence of non-homologous end joining.

RNA binder 1 (Compound 4b) is a blood-brain permeable RNA binder. RNA binder 1 can selectively bind to the G-quadruplex structure of the G4C2 repeat sequence RNA of the C9orf72 gene. RNA binder 1 significantly reduces the levels of toxic polypeptides poly(GA) and poly(GP) produced by the G4C2 repeat sequence in amyotrophic lateral sclerosis (ALS) patient-derived cells. RNA binder 1 has no significant effect on the antisense polypeptide poly(PR), showing selectivity for sense RNA. RNA binder 1 can be used in the study of ALS and frontotemporal dementia (FTD) .

Thermostable T7 RNA Polymerase is a thermostable version of T7 RNA Polymerase (HY-E70090). Compared with T7 RNA Polymerase, it has high temperature resistance and stable activity. T7 RNA polymerase is a polymerase expressed by Escherichia coli from the RNA polymerase gene of T7 bacteriophage. T7 RNA polymerase is highly specific and involved in in vitro transcription (IVT) of mRNA. In the presence of Mg 2+, T7 RNA polymerase only uses the single-stranded or double-stranded DNA containing the T7 promoter sequence as a template, and uses NTP as a substrate to synthesize RNA complementary to the single-stranded DNA downstream of the promoter .

Human TLR7 mRNA encodes the human toll like receptor 7 (TLR7) protein, a member of the Toll-like receptor (TLR) family which plays a fundamental role in pathogen recognition and activation of innate immunity. TLR7 senses single-stranded RNA oligonucleotides containing guanosine- and uridine-rich sequences from RNA viruses, a recognition occuring in the endosomes of plasmacytoid dendritic cells and B cells.

Molecular Glues

Research Areas:

Z-NCTS is a small molecule that binds to mismatched RNA, recognizes the 5'-r (XGG)-3'/5'-r (XGG)-3' sequence, and acts as an RNA molecular glue. Z-NCTS induces the formation of tertiary structures in ribozymes and activates their RNA cleavage activity. As an RNA molecular glue, Z-NCTS enables small molecule-based strategies for regulating RNA structure and function. Z-NCTS binds to d (CGG)/d (CGG) double-stranded DNA, recognizes four guanine bases via hydrogen bonding, and stabilizes the double-stranded DNA. Z-NCTS binds to r (GGG)/r (GGG) double-stranded RNA and enhances the thermodynamic stability of the RNA duplex. Z-NCTS acts as a translation inhibitor by inducing the formation of stable hairpin structures in the 5'-UTR region of engineered mRNA, thereby blocking ribosomal access to the binding site .

β4-TAT is a kind of β-sheet peptide foldamer. β4-TAT is composed of two parts: the β4 sequence (derived from the β-sheet fragment of the RRM domain of fused in sarcoma (FUS)) and the TAT sequence (a nuclear localization signal peptide that helps the entire peptide penetrate the nuclear membrane). β4-TAT influences FUS aggregation by targeting its RNA recognition motifs (RRM). β4-TAT effectively induces FUS aggregation within cells, leading to the death of cancer cells. β4-TAT can be used for the study of FUS .
