4-Methylumbelliferyl oleate is a fluorogenic substrate for acid and alkaline lipases. 4-Methylumbelliferyl oleate is cleaved by lipases, liberating 4-Methylumbelliferyl (Ex/Em=320/450 nm).

Biological Activity
Guide (The following is our recommended solution. This solution is merely a guideline and should be modified according to your specific needs.)
1. Prepare the reaction system: Add the following components in sequence to the microplate or reaction tube:
50 μL 0.1 mM 4-MU oleate (substrate)
20 μL McIlvane buffer (pH 7.4)
5 μL sample solution to be tested (dissolved in an appropriate solvent)
2. Add the enzyme solution: Add 25 μL porcine pancreatic lipase to initiate the reaction.
3. Volume Adjustment: If necessary, add buffer solution or deionized water to replenish the total volume of the reaction to 100 μL.
4. Incubation: Place the reaction system at 37°C for 10 minutes.
5. Detection: Measure the fluorescence intensity of the released 4-methylumbelliferone (4-MU).

Emission wavelength: 450 nm
