BODIPY505/515 is a BODIPY dye. BODIPY dye is a small molecule dye with strong ultraviolet absorption ability, its fluorescence peak is relatively sharp, and the quantum yield is high. They are relatively insensitive to the polarity and pH of the environment and are relatively stable under different physiological conditions. Due to its structural asymmetry, BODIPY derives a variety of structural products. BODIPY lipid droplet dyes can well pass through the cell membrane into the cell, and localize the neutral lipids in the cell to specifically stain the lipid droplets, which can be used for labeling of live cells and fixed cells. Maximum excitation/emission wavelength: 505/515 nm.

    In Vitro

    Guide (The following is our recommended solution. This solution is merely a guideline and should be modified according to your specific needs.)
    1. Preparation of BODIPY505/515 working solution
    1.1 Preparation of storage solution
    Dilute 1 mg of BODIPY505/515 with 382 μL of anhydrous DMSO to prepare a 10 mM stock solution.
    1.2 Preparation of working solution
    Dilute the stored solution with preheated serum-free cell culture medium or PBS to prepare a 1-10 μM BODIPY505/515 working solution.
    Note: Please adjust the concentration of the BODIPY505/515 working solution according to the actual situation, and prepare it as needed.
    2. Cell Staining (Suspension Cells)
    2.1 Centrifuge to collect the cells, then add PBS for two washes, each for 5 minutes. The cell density is 1×106/mL
    2.2 Add 1 mL of BODIPY505/515 working solution and incubate at room temperature for 5 to 30 minutes.
    2.3 400 g, centrifuge for 3-4 minutes, then discard the supernatant.
    2.4 Wash the cells twice with PBS, each time for 5 minutes.
    2.5 After resuspending the cells in 1 mL of serum-free medium or PBS, observe them using a fluorescence microscope or a flow cytometer.
    3. Cell Staining (Monolayer Cells)
    3.1 Place the monolayer cells on a sterile cover slip.
    3.2 Remove the cover glass from the culture medium and aspirate away the excess medium.
    3.3 Add 100 μL of the dye working solution and gently shake to ensure complete coverage of the cells. Incubate for 5 to 30 minutes.
    3.4 Remove the dye working solution and wash with the culture medium 2-3 times, each time for 5 minutes. Observe using a fluorescence microscope or flow cytometer.

    Notes
    1. Please adjust the concentration of BODIPY505/515 working solution and the incubation time according to the actual situation.
    2. It is recommended to conduct a positive control in the experiment. The cells in the control group should be incubated with 30 μM oleic acid for 8 hours before proceeding with the subsequent experiments.
