1. Membrane Transporter/Ion Channel
  2. SLC39 (Zinc Transporter)
  3. Anti-ZnT8 Antibody (mAb43)

Anti-ZnT8 Antibody (mAb43) is a monoclonal antibody targeting the zinc transporter ZnT8, with islet-specific biodistribution characteristics. Anti-ZnT8 Antibody (mAb43) binds to extracellular ZnT8 on the surface of pancreatic β-cells and masks its insulin-co-localizing sites to block autoimmune recognition. Anti-ZnT8 Antibody (mAb43) also promotes an increase in the proportion of regulatory T cells and inhibits B cell antigen presentation, thereby effectively blocking the T cell-mediated cascade of β-cell destruction. Anti-ZnT8 Antibody (mAb43) eliminates insulitis, preserves β-cell mass and induces seroconversion of autoantibodies, without directly altering the insulin secretion function or content of β-cells. Anti-ZnT8 Antibody (mAb43) can be used for research related to type 1 diabetes.

In Vivo

Anti-ZnT8 Antibody (mAb43) (5 mg/kg; i.v.; weekly; up to 25 weeks) administered from 10 weeks of age provides 100% sustained protection against spontaneous type 1 diabetes onset in female NOD mice through 35 weeks[1].
Anti-ZnT8 Antibody (mAb43) (5 mg/kg; i.v.; weekly; up to 26 weeks) administered from 14 weeks of age provides prolonged protection against spontaneous type 1 diabetes in female NOD mice[1].
Anti-ZnT8 Antibody (mAb43) (5 mg/kg; i.v.; weekly; 11 weeks) has no detectable adverse effects on β-cell function, glucose homeostasis, or insulitis in male B6 db mice with obesity-induced type 2 diabetes[1].

MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.

Animal Model: NOD/ShiLtJ (female, 10 weeks old at treatment initiation)[1]
Dosage: 5 mg/kg
Administration: i.v.; weekly; up to 25 weeks
Result: Maintained euglycemia through 35 weeks of age in all 64 treated mice, with only one mouse experiencing a transient hyperglycemic episode that resolved to FBG <250 mg/dL.
Achieved 100% nondiabetic rate, compared to 0% of control mice.
Exhibited significantly higher β-cell mass than diabetic control mice at all ages.
Showed median insulitis severity between that of control mice with acute or progressive T1D onset at 20 weeks of age; extending treatment to 35 weeks resulted in significant clearance of destructive insulitis.
Achieved complete insulin autoantibody (IAA) negativity by 35 weeks of age.
Significantly increased the number of CD4+FoxP3+CD25+ regulatory T cells (Tregs) within pancreatic islets, with no significant changes in total CD4+ T cells.
Shifted islet-residing CD4+ T cells toward higher FoxP3 expression without altering immune cell subsets or phenotypes in pancreatic lymph nodes or spleens.

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
