AMG 1541
AMG 1541 is a degradable ionizable lipid. AMG 1541 mediates the delivery of polynucleotides including mRNA. AMG 1541 exerts potent neutralizing effects on pseudotyped lentiviruses carrying the B.1.617.2 spike protein. As a component of influenza vaccines, AMG 1541 enables robust immune activation and reduces vaccine dosage. AMG 1541 is applicable to research related to SARS-CoV-2 infection.

Actividad biológica
Descripciòn
In Vitro
AMG 1541 potently neutralizes B.1.617.2 spike protein pseudovirus in 293T-ACE2 cells[1].
AMG 1541 (40 ng/well) mediates luciferase reporter gene expression in C2C12 cells, producing a detectable luminescent signal at a dose of 40 ng/well[1].
MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.
In Vivo
AMG 1541 (intravenous injection) effectively delivers mRNA to the liver of mice, achieving a total hepatic flux of approximately 1.1×1010 p/s[1].
AMG 1541 (0.25-1 mg/kg; intravenous injection) effectively delivers hEPO mRNA in mice, inducing a dose-dependent increase in serum hEPO levels, with an average level of 1200-1300 IU/mL (upper limit of approximately 1500 IU/mL) at the 1 mg/kg dose[1].
AMG 1541 (0.5-3 mg/kg; intravenous administration) mediates dose-dependent PCSK9 gene editing in mouse livers, achieves approximately 31% indels at the dose of 3 mg/kg, and reduces serum PCSK9 levels to approximately 5% of the baseline value[1].
MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.

Result:Achieved a mean log[Endpoint Titer] of approximately 6.7 in male mice and approximately 7.1 in female mice.
Induced significantly higher log[Endpoint Titer] (mean ~6.8) compared to SM-102 (mean ~5.9).
Induced a mean of ~2000 SFU/5×105 cells (range ~1000-2600) for MHC-I responses.
Induced a mean of ~500 SFU/5×105 cells (range ~0-700) for MHC-II responses.
Induced log[HAI Titer] values of ~2.5 at 0.01 µg, ~2.2 at 0.1 µg, and ~3.2 at 1 µg.
