Background

The Ephrin-A2/EFNA2 protein, a cell surface GPI-bound ligand for Eph receptors, is integral to neuronal, vascular, and epithelial development, where Eph receptors play a crucial role in migration, repulsion, and adhesion. EFNA2 binds promiscuously to Eph receptors on adjacent cells, initiating contact-dependent bidirectional signaling, with forward signaling downstream of the receptor and reverse signaling downstream of the ephrin ligand. In association with the EPHA2 receptor, EFNA2 may contribute to bone remodeling by regulating osteoclastogenesis and osteoblastogenesis.
