PTH (1-34) amide human is a type 1 PTH/PTHrP receptor agonist. PTH (1-34) amide human activates adenylate cyclase and phospholipase C pathways, thereby mediating mineral ion homeostasis and bone metabolism regulation. PTH (1-34) amide human increases serum calcium, decreases serum phosphorus, regulates renal excretion, while inducing the inhibition of endogenous PTH (1-84) and the increase of 1,25-dihydroxyvitamin D2 and bone resorption. PTH (1-34) amide human stimulates phosphatidylcholine hydrolysis via phospholipase D mediation, and its hypercalcemic effect is inhibited by human PTH-(7-84). PTH (1-34) amide human can be used in the research of diseases related to humoral hypercalcemia of malignancy .

1α-Hydroxyvitamin D5 (CARD-024) is a vitamin D analog with extremely low hypercalcemic effects. 1α-Hydroxyvitamin D5 effectively attenuates the pro-fibrotic response of colonic myofibroblasts to TGFβ and high-stiffness substrates by inhibiting the expression of αSMA, the phosphorylation of FAK, and the gene expression of MLCK and ET-1, while activating the anti-inflammatory pathway of IL-1β/COX-2. 1α-Hydroxyvitamin D5 can be used in research related to intestinal fibrosis associated with Crohn's disease .

A-75943 is a bone resorption inhibitor isolated from Streptomyces sp. SANK 61296, with protein synthesis toxicity. A-75943 inhibits bone resorption in vitro, and also suppresses the increase in serum calcium levels and the decrease in bone mineral density in thyroparathyroidectomized rats. A-75943 inhibits protein synthesis. High concentrations of A-75943 reduce the survival rate of osteoclasts on bone slices, while low concentrations have no effect on the survival rate or morphology of osteoclasts. A-75943 can be used for the research of osteoporosis and hypercalcemia .
