Biological Activity
Sphingomyelin phosphodiesterase from *Bacillus cereus* (0-200 μM; 30 min) hydrolyzes sphingomyelin in a dose-dependent manner, with a dissociation constant (Kd) of approximately 50 μM[1].
Sphingomyelin phosphodiesterase from *Bacillus cereus* (0.1-1.0 μg/mL; 60 min pre-incubation) dose-dependently inhibits peptidoglycan-induced H2O2 production and phagocytosis in mouse peritoneal macrophages, without altering TNF-α release or inducing cytotoxicity[3].
Sphingomyelin phosphodiesterase, Bacillus cereus (0.05-1.0 μg/mL; 60 min) dose-dependently increases ceramide levels in mouse peritoneal macrophages, induces the formation of ceramide-enriched membrane domains, and reduces membrane fluidity[3].
Sphingomyelin phosphodiesterase from *Bacillus cereus* (30 min) exhibits hemolytic activity against sheep red blood cells[3].

Recombinant *Bacillus cereus* sphingomyelin phosphodiesterase (administered via lateral injection, single dose) exhibits potent acute insecticidal activity against black cutworm larvae, with an MPD of 110 ng per insect; whereas inactivation of sphingomyelinase activity via the His151Ala mutation completely abolishes this toxicity[1].
Sphingomyelin phosphodiesterase from *Bacillus cereus* (0.1-5.0 μg per mouse; single dose) dose-dependently enhances the virulence of non-pathogenic B. cereus ATCC21928 in mice. At a dose of 5.0 μg per mouse, it induces 100% mortality, with a blood bacterial count of 300-400 CFU/100 μl[3].
Sphingomyelin phosphodiesterase, *Bacillus cereus* (a mixture of 25 μg Sphingomyelin phosphodiesterase, *Bacillus cereus* and complete Freund's adjuvant; administered subcutaneously; once every two weeks for a total of 2 times) provides complete protection to BALB/c mice against challenge with a lethal clinical isolate of B. cereus, resulting in a 100% survival rate of the mice[3].
