Guide (The following is our recommended protocol. This protocol is only a guide and should be modified according to your specific needs).
Reagent Preparation: Prepare a 10 g/L 2,4,6-Tris(2-pyridyl)-1,3,5-triazine (TPTZ) solution by dissolving TPTZ in a small amount of hydrochloric acid or ethanol.
Sample Preparation: Mix the iron-containing sample with a reducing agent (e.g., 1 mL of 100 g/L ascorbic acid) to ensure all iron is in the Fe(II) state.
Complexation: Add 1 mL of the TPTZ solution and 1 mL of an appropriate buffer (e.g., 1 M ammonium acetate, pH 5.4).
Measurement: Dilute to volume (e.g., 10 or 25 mL), allow it to stand for color development (approx. 10-30 min), and measure the absorbance of the blue [Fe(TPTZ)?]2? complex at 593 nm.
