Thiazolyl Blue  (Synonyms: MTT; Thiazolyl Blue Tetrazolium bromide; Methylthiazolyldiphenyl-tetrazolium bromide)

Cat. No.: HY-15924 Purity: 99.84%
Handling Instructions Technical Support

Thiazolyl Blue (MTT) is a cell-permeable and positively charged tetrazolium dye that is used to detect reductive metabolism in cells. Thiazolyl Blue is taken up by cells through the plasma membrane and then reduced to formazan by intracellular NAD (P) H-oxidoreductases. Thiazolyl Blue is frequently used in colorimetric assays to measure cell proliferation, cytotoxicity, and apoptosis.

In Vitro

Guide (The following is the experimental plan we recommend. This plan serves only as a reference guide. The specific operations should be adjusted according to your actual needs.)
1. Preparation of MTT solution
Dissolve MTT in anhydrous DMSO to obtain a 5 mg/mL solution.
2. Cell proliferation assay (96-well plate)
2.1 Cell inoculation: Prepare single cells and prepare a suspension with 10% FBS culture medium. Inoculate them into a 96-well plate, with 1000-10,000 cells per well, and each well volume is 100 μL.
2.2 Cell culture: At 37°C, with 5% CO2, incubate for 24-72 hours.
2.3 Add 10 μL of MTT to each well, incubate for 4 hours, discard the supernatant, and the cells need to be centrifuged first.
2.4 Add 100 μL of DMSO, shake for 10 minutes to completely dissolve the crystals.
2.5 Use an absorbance plate reader to monitor the increase in absorbance at OD = 562 nm.

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
