AMPK α2β2γ1

AMPK α2β2γ1 belongs to the AMPK αβγ heterotrimer family that maintains cellular energy homeostasis through isoform-specific complexes[1]. Mechanistically, α2β2γ1 is relevant to skeletal muscle metabolism because human skeletal muscle contains α1β2γ1, α2β2γ1, and α2β2γ3 complexes, but high-intensity exercise selectively activates α2β2γ3 rather than α2β2γ1[2]. Compared with γ3-containing AMPK, γ1 predominates in stabilizing the AMPKα2β2γ1 complex, yet γ1 is dispensable for AICAR- or MK-8722-stimulated glucose uptake in skeletal muscle models[1]. For experimental applications, ADaM-site activators such as PF739 and 991 increase skeletal muscle glucose uptake independently of AMPKγ3, and PF739 selectively activates muscle AMPKγ1 complexes in vivo[3]. SC4 provides a direct α2-focused tool because it preferentially activates α2 complexes, stimulates skeletal muscle glucose uptake, and binds activated α2β2γ1 in structural studies[4].
