Antibiofilm agent-21 is an orally active prodrug of Antibiofilm agent-20 (HY-182268) with glucuronidation. Antibiofilm agent-21 avoids absorption in the small intestine via reduced membrane permeability, improving colon delivery rate, and converts to its active form in the colon. Antibiofilm agent-21 is an orally active prodrug of Antibiofilm agent-20 (HY-182268) with glucuronidation. In Vitro

Antibiofilm agent-21 (Compound 3) weakly inhibit Escherichia coli biofilm formation (IC50 >100 μg/mL) and exhibits antibacterial activity against Escherichia coli (MIC >128 μg/mL)[1].
Antibiofilm agent-21 has improved solubility, very low membrane permeability, and high metabolic stability (82% remaining after 30 min in mouse liver microsomes)[1].

In Vivo

Antibiofilm agent-21 (Compound 3) (30-100 mg/kg; p.o.; once daily; 7 consecutive days) significantly increases colon length and reduces colonic inflammatory cytokine levels in a murine DSS (HY-116282C)-induced colitis model[1].

Dosage: 30 mg/kg; 100 mg/kg

Result: Increased colon length to a mean of ~8 cm, with a statistically significant difference relative to the control group.
Reduced colon tissue IL-17 levels to a mean of ~50 pg/g tissue, with a statistically significant difference relative to the control group.
