Carbonic anhydrase-IN-39 is a bacterial carbonic anhydrase inhibitor with oral bioavailability. Carbonic anhydrase-IN-39 inhibits α-NgCA and β-NgCA from Neisseria gonorrhoeae, as well as human hCA I and hCA II. Carbonic anhydrase-IN-39 exhibits bactericidal activity against Neisseria gonorrhoeae and reduces gonococcal burden in infected mouse models. Carbonic anhydrase-IN-39 can be used in the research of gonorrhea.

Description

Carbonic anhydrase-IN-39 is a bacterial carbonic anhydrase inhibitor with oral bioavailability. Carbonic anhydrase-IN-39 inhibits α-NgCA and β-NgCA from Neisseria gonorrhoeae, as well as human hCA I and hCA II. Carbonic anhydrase-IN-39 exhibits bactericidal activity against Neisseria gonorrhoeae and reduces gonococcal burden in infected mouse models. Carbonic anhydrase-IN-39 can be used in the research of gonorrhea[1].

In Vitro

Carbonic anhydrase-IN-39 (Compound 18) inhibits the growth of drug-resistant *Neisseria gonorrhoeae* strains CDC 178 and CDC 181 under ambient air conditions, but loses its activity in a 5% CO2 environment[1].
Carbonic anhydrase-IN-39 inhibits purified *Neisseria gonorrhoeae* α-NgCA (Ki=35.4 nM), β-NgCA (Ki=660 nM), human hCA I (Ki=60.4 nM), and human hCA II (Ki=5.3 nM) in CO2 hydration stopped-flow assays[1].
Carbonic anhydrase-IN-39 inhibits the growth of clinically isolated multidrug-resistant *Neisseria gonorrhoeae* strains under ambient air conditions[1].
Carbonic anhydrase-IN-39 (1 μM; 0-24 h) exhibits bactericidal activity against *Neisseria gonorrhoeae* WHO-X, reducing the bacterial load by approximately 4.0 Log10 CFU/mL[1].

In Vivo

Carbonic anhydrase-IN-39 (20 mg/kg, p.o., two days post-infection, twice daily, three consecutive days) can significantly reduce the bacterial load of Neisseria gonorrhoeae in a mouse Gonococcal infection model[1].

MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.

Animal Model: BALB/c mice infected
intravaginally with N. gonorrheae WHO-X[1]
Dosage: 20 mg/kg
Administration: p.o., two days post-infection, twice daily, three consecutive days
Result: Had an approximately 1.5-log10 (96.84%) CFU/mL reduction in
gonococcal burden compared to vehicle-treated mice.

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
