Biological Activity
The STEAP1−3a-D2 PAC with a DAR of 1.7, constructed using cBu-Cit-GAL-02-221 (cBu-Cit-PROTAC BRD4 Degrader-5), degrades BRD4 in STEAP1-overexpressing PC3-S1 cells after 72 h of treatment. The absolute DC50 of the conjugate is 48 μg/mL, the DC50 calculated based on payload equivalent is 556 nM, and the maximum reduction level of BRD4 reaches 55%[1].
The DAR 5.7 STEAP1−3a-D6 PAC constructed from cBu-Cit-GAL-02-221 degrades BRD4 in PC3-S1 cells after 72 h of treatment. Its absolute DC50 value is 0.21 μg/mL, the DC50 calculated on a payload equivalent basis is 7.9 nM, and the maximum reduction level of BRD4 reaches 70%[1].
