PEAK

PEAK proteins (PEAK1/SGK269, PEAK2/SGK223, and PEAK3) form a family of large cytoplasmic pseudokinases whose kinase-related domains lack catalytic activity yet serve as regulated signaling scaffolds. PEAK1 localizes to focal adhesions, binds Grb2 and 14-3-3 proteins, and modulates integrin, ERK, and SRC signaling that drives cell migration and proliferation. PEAK members are frequently overexpressed in cancers, including breast and colorectal tumors, where they support metastatic behavior and resistance to targeted agents. Because they act as signaling hubs rather than enzymes, they are attractive but challenging drug targets. Recombinant PEAK proteins are used for interaction studies, antibody generation, and phosphoproteomic work.
