TP53RK
TP53RK, also called PRPK (p53-related protein kinase), is an atypical serine/threonine kinase of the Bud32 family whose transcription is induced by p53 following DNA damage. In cells, TP53RK assembles with OSGEP, LAGE3 and TPRKB into the KEOPS/EKC complex, which catalyzes the N6-threonylcarbamoyladenosine (t6A) modification of a subset of tRNAs and thereby supports translation fidelity and protein homeostasis; the complex also contributes to telomere maintenance in yeast and human cells. Because p53 upregulates TP53RK, the kinase is thought to modulate p53-dependent checkpoint signaling, although its direct substrates remain poorly defined. Recombinant TP53RK is useful for studying KEOPS complex assembly, t6A tRNA modification and p53 signaling crosstalk.
