TRRAP

null TRRAP is a catalytically dead member of the PIKK family: although it retains a kinase-like fold, key catalytic residues are absent, and it functions instead as an essential scaffold. It is the common core subunit of the TIP60/NuA4 histone acetyltransferase complex and of SAGA-related complexes, coupling transcription factors such as MYC, E2F and p53 to chromatin acetylation, DNA repair and mitotic progression. TRRAP is indispensable for cell viability and for MYC-driven transformation, and its depletion destabilizes both TIP60/NuA4 and SAGA assemblies. Because it lacks enzymatic activity, recombinant TRRAP is used for complex reconstitution, interaction mapping and studies of chromatin-signaling assembly rather than for kinase assays.
