Description

METTL3-IN-13 is a METTL3 inhibitor. METTL3-IN-13 is applicable to the research of multiple cancers such as hypopharyngeal squamous cell carcinoma and non-small cell lung cancer[1].

In Vitro

METTL3-IN-13 (Compound 142) (72 h) potently inhibits the viability of Calu-6, A549, Fadu, HCT-116, MDA-MB-231, PANC-1, 5637, DU 145 and PC-3, with an IC50 ≤ 200 nM[1].

In Vivo

METTL3-IN-13 (Compound 142) (30 mg/kg, i.p., once daily for 20 consecutive days) significantly inhibits subcutaneous hypopharyngeal squamous cell carcinoma tumor growth and prolongs survival in female NSG mice[1].
METTL3-IN-13 (30 mg/kg, i.p., once daily for 50 consecutive days) significantly prolongs the survival of female C.B-17 SCID mice in an orthotopic non-small cell lung adenocarcinoma model[1].

Animal Model: NOD.Cg-Prkdcscid Il2rgtm1Wjl/SzJ (NSG) (female, 6-7 weeks old, subcutaneous FaDu human hypopharyngeal squamous cell carcinoma implantation)[1]
Dosage: 30 mg/kg
Administration: i.p.; daily; day 5 to day 24
Result: Reduced day 24 tumor volume to ~800 mm3 (vs ~2000 mm3 in vehicle control).
Prolonged survival: 80% survived to day 32, and 20% survived to day 40 (all vehicle-treated mice died by day 24).
Animal Model: C.B-17 SCID (C.B-17/IcrHsd-Prkdcscid) (female, 6-7 weeks old, orthotopic A549-Luc-C8 human non-small cell lung adenocarcinoma implantation)[1]
Dosage: 30 mg/kg
Administration: i.p.; daily; day 16 to day 66
Result: Prolonged survival: 50% survived to day 60, and 10% survived to day 75 (all vehicle-treated mice died by day 45).
