    Description

NAMPT protein catalyzes the synthesis of nicotinamide mononucleotide, a key intermediate in NAD biosynthesis. NAMPT Protein, Human (His) is the recombinant human-derived NAMPT protein, expressed by E. coli , with N-6*His labeled tag.

    Background

NAMPT serves as the catalyst for the condensation reaction between nicotinamide and 5-phosphoribosyl-1-pyrophosphate, giving rise to nicotinamide mononucleotide—a pivotal intermediate in the biosynthesis of NAD. As the rate-limiting factor in the mammalian NAD biosynthetic pathway, NAMPT holds a central role in cellular processes. Beyond its enzymatic function, the secreted form of NAMPT exhibits dual properties, acting as both a cytokine with immunomodulating effects and an adipokine with anti-diabetic attributes. Notably, its extracellular function lacks enzymatic activity, partially due to the absence of activation by ATP, which is present at low levels in the extracellular space and plasma. Moreover, NAMPT plays a crucial role in the modulation of circadian clock function, orchestrating the oscillatory production of NAD to regulate clock target gene expression by facilitating the release of the CLOCK-BMAL1 heterodimer from NAD-dependent SIRT1-mediated suppression.

    Biological Activity

1.Measured by its ability to convert PRPP to NMN. The specific activity is 15.13987 pmol/min/µg, as measured under the described conditions.
2.Measured by its ability to induce VEGF secretion by PC-3 human prostate cancer cells. The ED50 for this effect is 0.5-2.0 μg/mL.

    • Measured by its ability to induce VEGF secretion by PC-3 human prostate cancer cells. The ED50 for this effect is 0.7365 μg/mL, corresponding to a specific activity is 1.36×103 U/mg.

    Reconstitution

It is not recommended to reconstitute to a concentration less than 100 μg/mL in ddH2O. For long term storage it is recommended to add a carrier protein (0.1% BSA, 5% HSA, 10% FBS or 5% Trehalose).

    Storage & Stability

Stored at -20°C for 2 years from date of receipt. After reconstitution, it is stable at 4°C for 1 week or -20°C for longer (with carrier protein). It is recommended to freeze aliquots at -20°C or -80°C for extended storage.

    Help & FAQs
    • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.

    • Specific Activity Calculator

    The reconstitution calculator equation

Volume (to add to vial) = Mass (in vial) ÷ Desired Reconstitution Concentration

Volume (to add to vial) = Mass (in vial) ÷ Desired Reconstitution Concentration
    = ÷

    The dilution calculator equation

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)

This equation is commonly abbreviated as: C1V1 = C2V2

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)
    × = ×
    C1   V1   C2   V2

    The specific activity calculator equation

    Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)

    Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)
    Unit/mg = 106 ÷ ng/mL
