Volrustomig (MEDI-5752) is a human IgG1 κ monoclonal antibody targeting CTLA4/PD1. The isotype control for Volrustomig is Human IgG1 kappa, Isotype Control (HY-P99001). Volrustomig anchors to the surface of T cells by binding PD-1, induces PD-1 internalization and degradation, and preferentially inhibits CTLA-4 on activated PD-1 + T cells. Volrustomig binds to tumor-infiltrating lymphocytes and a subset of PD-1 + B cells, enhances T cell function and IFNγ secretion. Volrustomig reduces the activation of non-tumor-infiltrating lymphocytes and exhibits manageable toxicity. Volrustomig can be used in research on various cancers, such as non-small cell lung cancer, gastric cancer, hepatobiliary cancer, and cervical cancer .

Cys(Npys)-(Arg)9 is a synthetic cationic cell-penetrating peptide with a reversible thiol-reactive nitropyridyl (Npys) group. Cys(Npys)-(Arg)9 efficiently mediates the internalization and delivery of various "cargo" such as proteins and antibodies by forming reversible disulfide bonds with surface-exposed cysteine residues. Cys(Npys)-(Arg)9 endows TALEN proteins with cell-penetrating activity, enabling gene knockout in mammalian cells and protein transduction in wheat microspores. Cys(Npys)-(Arg)9 can be conjugated with antibodies to form cationized IgG for enhancing endosomal escape of oligonucleotides, or form siRNA delivery complexes. When the molar ratio of Cys(Npys)-(Arg)9 to loaded molecules is higher than 1:1, it exerts certain cytotoxic effects on cells. Cys(Npys)-(Arg)9 can be used in studies related to oral squamous cell carcinoma and HIV infection .

Cyclazosin is an α1D-adrenergic receptor antagonist and a partial agonist of CXCR4/ACKR3. The pKi values of Cyclazosin for cloned human α1D, α1B and α1A adrenergic receptors are 9.28, 9.23 and 8.18, respectively. Cyclazosin induces β-arrestin recruitment in CXCR4 and ACKR3 with EC50 values of 16 μM and 10 μM, respectively, stimulates ERK1/2 phosphorylation, and induces receptor internalization. Cyclazosin potently inhibits CXCL12-induced chemotaxis of primary human aortic vascular smooth muscle cells. Cyclazosin alters MDMA-induced thermoregulatory responses in mice, converting monophasic hyperthermia to a biphasic pattern without affecting resting core body temperature. Cyclazosin can be used for diseases related to tumor metastasis, MDMA-induced hyperthermia and vasoconstriction .

ZW-191 is an antibody-drug conjugate (ADC) targeting folate receptor α (FRα). ZW-191 consists of ZW-191 Antibody (HY-P992515) as the antibody component and MC-GGFG-AM-(10Me-11F-Camptothecin) (HY-153360) as the linker-payload component. ZW-191 possesses excellent tumor sphere penetration, cellular internalization and payload delivery capabilities, and can release a topoisomerase 1 inhibitor inside tumor cells to inhibit tumor cell activity. ZW-191 exerts a potent bystander effect on both antigen-positive and antigen-negative cells. ZW-191 exhibits prominent activity in xenograft models of cell lines with different FRα expression levels. ZW-191 can be used in studies related to ovarian cancer, non-small cell lung cancer, endometrial cancer and triple-negative breast cancer .

AZD5335 is an ADC targeting FRα, with an IC50 of 10.6 nM against human FRα. AZD5335 specifically binds to FRα, mediates internalization and trafficking to lysosomal compartments, while delivering a topoisomerase I inhibitor payload that traps TOP1-DNA cleavage complexes. AZD5335 induces DNA damage response via the ATR, ATM, Chk1 and Chk2 signaling pathways, and increases the levels of phosphorylated KAP1, phosphorylated RPA, phosphorylated γH2AX, as well as PARP cleavage. AZD5335 exhibits bystander cytotoxicity against FRα-negative cells co-cultured with FRα-expressing cells. AZD5335 induces potent and durable responses and complete regression in ovarian cancer models, including models with low FRα expression and models resistant to Elahere, with enhanced efficacy when combined with Carboplatin, Bevacizumab or poly (ADP-ribose) polymerase inhibitors. AZD5335 can be used for ovarian cancer research .

Gestodene (SHB 331; WL 70) is an orally active synthetic progestogen compound of the 19-nortestosterone class. Gestodene binds with high affinity to the progesterone receptor, inhibits 5α-reductase, binds to androgen and aldosterone receptors, and inactivates CYP3A. Gestodene acts as a positive allosteric modulator of PAR1, enhances PAR1-mediated signaling pathways, and is capable of increasing the activation potency of PAR1-AP toward PAR1, thereby promoting ERK1/2 phosphorylation, receptor internalization, cell morphological changes, and human platelet aggregation. Gestodene and its A-ring reduced metabolites promote the proliferation, differentiation, and mineralization of neonatal rat osteoblasts. Gestodene itself has no binding capacity for estrogen receptors, and this osteogenic activity depends on intracellular metabolism to generate A-ring reduced products with estrogen-like agonistic activity. Gestodene is useful for research on diseases related to progesterone secretion and diseases related to thrombosis .

Ro 106-9920 is an orally active NF-κB inhibitor. Ro 106-9920 prevents ubiquitination of IκBα, blocks nuclear translocation of NF-κB, and inhibits NF-κB activation. Ro 106-9920 suppresses TNF-α-induced tissue factor expression and procoagulant activity, enhances TNF-α-mediated cytotoxicity against cancer cells, and eliminates the formation of neutrophil extracellular traps. Ro 106-9920 inhibits NLRP3 inflammasome activation, reduces inflammatory cytokine secretion, decreases myeloperoxidase activity, attenuates renal cell apoptosis, and improves renal function. Fluorescent Dye

Research Areas:

Infection

CY5-Dextran (MW60-80k) is a fluorescent probe formed by covalent conjugation of the cyanine dye Cy5 to dextran. It is a pH-insensitive fluid-phase endocytosis tracer (the pH of endosomal lumen is precisely quantified by the fluorescence ratio of FITC/Cy5). CY5-Dextran (MW60-80k) is internalized by cells via a clathrin-independent fluid-phase pinocytosis pathway, and is sequentially evidenced by early endosomes, late endosomes are transported to lysosomes. CY5-D I cannot pass through the limited pore size formed by uncoating of human rhinovirus type 2 (HRV2), but can be released into the neutral cytoplasm upon adenovirus-mediated endosomal rupture, thus distinguishing between the two viral modes of "pore-forming escape" and "lysis escape". CY5-Dextran (MW60) -80k) has an excitation wavelength of 635 nm, and it can also be used for time-lapse imaging of endocytosis dynamics of stromal cells in tissues such as rat salivary glands under in vivo two-photon microscopy .
