ZW-191 is an antibody-drug conjugate (ADC) targeting folate receptor α (FRα). ZW-191 possesses excellent tumor sphere penetration, cellular internalization and payload delivery capabilities, and can release a topoisomerase 1 inhibitor inside tumor cells to inhibit tumor cell activity. In Vitro

ZW-191 shows detectable apparent binding affinity in FRα-expressing cells. ZW-191 mediates superior cellular internalization and payload delivery compared with other FRα-targeted antibodies applied in different ADCs[1].
ZW191 demonstrates strong FRα binding, superior tumor spheroid penetration, cellular internalization, and cytotoxic activity in FRα-expressing cancer cells[1][2].

In Vivo

ZW-191 exerts potent antitumor activity in cell line-derived xenograft and patient-derived xenograft models covering different FRα expression levels. ZW-191 maintains favorable tolerability in rodents after two administrations at a dose of 200 mg/kg. ZW-191 exhibits satisfactory safety profiles in non-human primates when given twice at a dose of 30 mg/kg. ZW-191 displays optimal pharmacokinetic characteristics in rodents and non-human primates during in vivo assessments[1].
