Function
OTULIN is a deubiquitinase that specifically cleaves linear ('Met-1'-linked) polyubiquitin chains and plays a critical role in angiogenesis, innate immunity, and the regulation of Wnt and NF-kappa-B signaling pathways. It supports angiogenesis, craniofacial, and neuronal development by modulating canonical Wnt signaling alongside the LUBAC complex. As a negative regulator of NF-kappa-B, OTULIN controls LUBAC activity by removing its autoubiquitination, thus maintaining its function. It also acts as a key suppressor of inflammation and autoimmunity by restricting excessive cytokine secretion and linear ubiquitination. During NOD2 stimulation, OTULIN limits linear ubiquitin chain formation on LUBAC to modulate innate immune responses. by similar: OTULIN primarily functions in maintaining LUBAC complex homeostasis.
