JSD26
JSD26 is a selective and orally active SCoR2 (AKR1A1) inhibitor with an IC50 of 93 nM. JSD26 has 10-fold selectivity for SCoR2 versus AKR1B1. JSD26 can be used for the study acute kidney injury (AKI).

JSD26 (120 mg/kg, i.p., once) alleviates symptoms in acute kidney injury (AKI) mice by selectively inhibiting SCoR2 and increasing S-nitrosylation of PKM2[1].

Result:At the highest dose (120 mg/kg), renal SCoR2 activity was inhibited by 80%.
Did not inhibit renal AKR1B1 activity and inhibited SCoR2 in other organs (liver, lung, heart, spleen), but showed no inhibition in brain tissue, indicating that it cannot cross the blood-brain barrier.

Result:Renal SCoR2 activity was inhibited by 80%, but SCoR2 protein expression levels remained unchanged.
Serious creatinine and BUN levels were significantly reduced, indicating improved renal function.
Histological analysis (H&E staining) showed reduced tubular damage in the JSD26 treatment group (e.g., reduced tubular dissolution and debris).
PKM2 S-nitrosylation (SNO-PKM2) levels were increased, and PKM2 activity was decreased.
