MCE Centrifugal Protein Desalting Column (5 kDa) is a prepacked desalting device containing resin particles with a particle size of 57–80 μm. It efficiently separates small molecules with molecular weights ≤ 5 kDa (such as salts and buffer components) from macromolecules ≥ 5 kDa (such as proteins and peptides), making it suitable for protein desalting and buffer exchange applications.

MCE Centrifugal Protein Desalting Column (5 kDa) is a prepacked desalting device containing resin particles with a particle size of 57–80 μm. It efficiently separates small molecules with molecular weights ≤ 5 kDa (such as salts and buffer components) from macromolecules ≥ 5 kDa (such as proteins and peptides), making it suitable for protein desalting and buffer exchange applications.
This product achieves a retention rate of more than 95% for salts and small molecules. During desalting or buffer exchange, it enables nearly complete volume recovery without significant sample dilution while maintaining high protein recovery rates. The prepacked format eliminates the need for column packing and serves as a rapid alternative to traditional dialysis methods. With only simple centrifugation steps, protein desalting or buffer exchange can be completed within a short time, making it ideal for rapid protein purification and downstream sample preparation.

Protocols
Column Preparation
1. Remove the Centrifugal Protein Desalting Column (5 kDa) from the package and carefully remove the bottom sealing plug and top cap.
2. Place the column vertically into the corresponding collection tube.
Column Equilibration
1. Add PBS to fully wet and swell the prepacked resin. As a reference from MCE's related 5 kDa rapid desalting column, use 6 mL PBS for a 1 mL format or 600 μL for a 250 μL format and allow the column to stand for approximately 3 min at room temperature.
2. Centrifuge at approximately 1200 × g for 3 min and discard the filtrate.
3. Repeat PBS equilibration twice, keeping the resin-bed surface undisturbed and level.
Sample Desalting / Buffer Exchange
1. Apply the protein or peptide sample evenly to the center of the resin bed and avoid disturbing the bed surface.
2. Centrifuge to separate target macromolecules ≥5 kDa from salts, free dyes, and other small molecules ≤5 kDa.
3. Collect the desalted target-protein fraction. For further buffer exchange, re-equilibrate and repeat using the desired buffer.
Note: If the sample concentration is too low, concentrate the sample to a suitable volume or concentration before desalting. The column is intended for rapid removal of small-molecule impurities and buffer exchange.

Attention
1. This product is recommended for single use, but it may also be regenerated and reused.
2. Recommended operating conditions: pH range 2–13; operating temperature 4–30°C.
3. Strictly follow the recommended sample loading range. Excessive sample loading may result in incomplete desalting, whereas insufficient loading may reduce protein recovery.
4. The resin may undergo dehydration and shrinkage in high-concentration alcohol solutions or saturated salt solutions. Direct application of such solutions onto the column should be avoided whenever possible.
5. When loading samples, apply the sample evenly to the center of the column bed to achieve optimal separation performance.
6. For low-concentration samples, concentration prior to desalting is recommended to improve recovery efficiency.
