Description

Lug-15 is a rapid bactericidal agent. Lug-15 exhibits strong antibacterial activity against both Gram-positive and Gram-negative bacteria, including drug-resistant strains. Lug-15 rapidly kills bacteria primarily through membrane disruption and had a very low propensity to induce bacterial resistance. Lug-15 demonstrates low hemolytic toxicity and significant therapeutic potential in various infection models. Lug-15 can be used for research on combating infections caused by multi-drug resistant bacteria[1].

In Vitro

Lug-15 exhibits potent antibacterial activity against both Gram-positive and Gram-negative bacteria tested with MICs of 3.13 μg/mL (S. aureus and Methicillin-resistant S. aureus (MRSA)), 6.25 μg/mL (E. gallinarum) and 12.5 μg/mL (P. aeruginosa, baumannii and E. coli)[1].
Lug-15 (2 × MIC, 0-12 h) reduces the S. aureus and E. coli count from 10^5.5 CFU/mL to 0 within 4 hours[1].
Lug-15 (× MIC, 0.5-1 h) disrupted the integrity of the bacterial outer membrane/membrane of S. aureus and E. coli and damaged the inner membrane of E. coli, causing irreversible damage to the cell membrane[1].
Lug-15 (0.5-4 × MIC, 4 h) significantly reduces the ATP content within bacterial cells and causes a concentration-dependent significant increase in ROS levels in S. aureus[1].
Lug-15 (0.5 MIC, passaged for 21 times) is extremely difficult to induce S. aureus to develop resistance[1].
Lug-15 (6.25-200 μg/mL, 1 h) exhibits the hemolysis rate in rat red blood cells was below 10% at the effective antibacterial concentration[1].
Lug-15 (0-48 h) has good stability in mammalian serum and can maintain its antibacterial activity in complex physiological environments[1].

In Vivo

Lug-15 (1 mg/mL, eye drops, multiple times) is effective in treating MRSA or P. aeruginosa infections in the eyes in mice[1].
Lug-15 (5-15 mg/kg, i.p. or local use of hydrogel, single dose or 2-4 doses) demonstrates strong efficacy in models of MRSA infections in the thigh, lungs, and wounds[1].

Animal Model: Keratitis Model infected with MRSA or P. aeruginosa established in six-week-old female Kunming mice (weighing 20-22 g)[1]
