Pentafluorobenzenesulfonyl fluorescein is a H2O2-selective sensor that can be used to detect H2O2 levels in cells. Pentafluorobenzenesulfonyl fluorescein is generally non-fluorescent, but emits fluorescence when its sulfonyl bond undergoes perhydrolysis by H2O2. Pentafluorobenzenesulfonyl fluorescein undergoes slight cleavage of its sulfonate ester bond by [Cu (phen)2]2+, and can detect hydrogen peroxide around the ablation sites of fin tissues and keratinocytes in zebrafish larvae.

Description

Pentafluorobenzenesulfonyl fluorescein is a H2O2-selective sensor that can be used to detect H2O2 levels in cells. Pentafluorobenzenesulfonyl fluorescein is generally non-fluorescent, but emits fluorescence when its sulfonyl bond undergoes perhydrolysis by H2O2. Pentafluorobenzenesulfonyl fluorescein undergoes slight cleavage of its sulfonate ester bond by [Cu (phen)2]2+, and can detect hydrogen peroxide around the ablation sites of fin tissues and keratinocytes in zebrafish larvae[1][2][3].

In Vivo

Pentafluorobenzenesulfonyl fluorescein (4µM; incubated in media; single 1-2 hour exposure prior to imaging) detects significantly increased hydrogen peroxide levels in the caudal fin of glucose-treated larval Danio rerio, with elevated regional fluorescence at 5dpf and widespread, intensified fluorescence at 8dpf[1].

Animal Model: Nacre, Tuebingen, Tg(isl2b:GFP), Tg(NF-κB:GFP) (larval stage, 2-8 days post fertilization)[1]
