Background

FOLR4, a receptor located at the oolemma (cell surface of oocytes), plays a pivotal role in species-specific gamete recognition and fertilization through its interaction with IZUMO1. This adhesion event is essential for successful fertilization, although it alone is not sufficient for cell fusion. Unlike its name might suggest, FOLR4 does not bind folate. Structurally, FOLR4 exists as a monomer and directly interacts with IZUMO1, forming a complex with a 1:1 stoichiometry. The IZUMO1:IZUMO1R/JUNO interaction mediated by FOLR4 is a crucial step in the intricate process of sperm and egg recognition, highlighting its significance in the fertilization cascade.
