Biological Activity

Description

IL-2 Protein, Human activates STAT, PI3K and MAPK pathways. IL-2 Protein, Human increases NK-cell cytolytic activity as a T-cell growth factor. IL-2 Protein, Human regulates the proliferation of activated B-cells and regulatory T-cells. IL-2 Protein, Human is the recombinant human-derived IL-2 Protein, expressed by E. coli.

Background

Recombinant human interleukin-2 (IL-2) expressed as Escherichia coli is isolated as insoluble aggregates of protein (inclusion bodies) after cell breakage. Interleukin 2 (IL-2), a cytokine secreted by activated T cells, promotes the differentiation and proliferation of lymphoid cells as well as enhances the cell-mediated immune response to infections. Therefore IL-2 has been used as an adjunctive immunotherapeutic agent to control some infectious diseases, such as leishmaniasis, leprosy, and HIV infection[1].

In Vitro

IL-2 Protein (Human, 500 U/mL, 42 hours-5 days) induces cytotoxic activity in mouse splenocytes and human peripheral blood lymphocytes, which exhibits a killing effect on tumor cells[2].

Verified Bioactivity

1. The ED50 as determined by the dose-dependant stimulation of the proliferation of CTLL-2 was found to be < 0.1 ng/ml, corresponding to a specific activity of > 1.0 × 107 IU/mg.
2. Measured in a cell proliferation assay using CTLL-2 mouse cytotoxic T cells. The ED50 for this effect is <3.0 ng/mL.

Results
  • Experimental Validation Results for IL-2 Protein, Human
    The ED50 is 0.1 ng/mL as measured by CTLL-2 cells, corresponding to a specific activity of 1 × 107 units/mg.

Reconstitution

It is not recommended to reconstitute to a concentration less than 100 μg/mL in ddH2O. For long term storage it is recommended to add a carrier protein (0.1% BSA, 5% HSA, 10% FBS or 5% Trehalose).

Storage & Stability

Stored at -20°C for 2 years from date of receipt. After reconstitution, it is stable at 4°C for 1 week or -20°C for longer (with carrier protein). It is recommended to freeze aliquots at -20°C or -80°C for extended storage.
