5-Monobutyl methotrexate (NSC 305986; γ-MBMTX) is a dihydrofolate reductase (DHFR) inhibitor and esterase substrate. 5-Monobutyl methotrexate is hydrolyzed by esterases to produce methotrexate. In rhesus monkeys, 5-Monobutyl methotrexate clears more slowly from cerebrospinal fluid than from serum, and its serum protein binding rate exceeds 99%. At the same dose rate, 5-Monobutyl methotrexate achieves cerebrospinal fluid concentrations comparable to those of Methotrexate (HY-14519), and exhibits activity against Methotrexate-resistant cells (with transport defects) that is comparable to that against sensitive parental cells. 5-Monobutyl methotrexate can be used in studies related to Methotrexate-resistant tumors .

BP Fluor 430 NHS Ester is a water-soluble, green-fluorescent dye that is often used in multi-color applications, including flow cytometry and super-resolution microscopy using STORM. Its excitation is ideally suited for the 407 nm spectral line of the krypton laser or the 408 nm violet laser diode. BP Fluor 405 conjugates are pH-insensitive from pH 4 to pH 10. The NHS ester (or succinimidyl ester) is the most popular amine reactive group for labeling the primary amines of proteins (Lys), amine-modified oligonucleotides, and other amine-containing molecules. Proteins can be labeled with BP Fluor 430 NHS Ester at high molar ratios without significant self-quenching, leading to brighter conjugates and more sensitive detection.

NtHzBtd is a fluorescent probe for detecting Fe 3+. NtHzBtd is applicable to the selective fluorescent detection of Fe 3+ ions and live cell imaging studies. NtHzBtd can selectively coordinate with Fe 3+ to form a 1:1 complex, triggering chelation enhanced quenching (CHEQ) and intramolecular charge transfer (ICT) processes, which result in fluorescence turn-off, thereby enabling sensitive detection of Fe 3+ and live cell fluorescence imaging. After binding to Fe 3+, NtHzBtd reduces fluorescence intensity, exhibits a rapid response property, with a limit of detection of 0.036 μM and a response time of approximately 55 s. The detection wavelengths are Ex/Em = 334/401 nm (solution system) .

Di-4-AN (F) EPPTEA is a voltage-sensitive dye used for membrane potential imaging and optical recording of electrical activity in excitable cells and tissues. Its detection mechanism is based on electrochromism (also known as the molecular Stark effect). The single-photon excitation/emission wavelengths measured in ethanol solution are Ex/Em = 444/610 nm, and it can be excited via 1064 nm two-photon excitation in optical recording applications. Di-4-AN (F) EPPTEA can be used to record backpropagating action potentials in individual dendritic spines of brain cortical slices, map subcellular action potential characteristics of single cardiomyocytes, and perform real-time recording of multicellular action potentials in cerebellar slices .

Di-4-ANEPPDHQ (solution) is a polarity-sensitive membrane probe with a doubly positive charge that can be used to visualize membrane microdomains in living Arabidopsis cells. Di-4-ANEPPDHQ produces a fluorescence spectrum shift by changing the polarity of the lipid environment after embedding into the cell membrane, thereby quantifying the degree of lipid accumulation. The excitation wavelength of Di-4-ANEPPDHQ is 488 nm, and the emission wavelengths are 560 nm and 650 nm in the liquid ordered and disordered phases, respectively. Di-4-ANEPPDHQ is often used for fluorescence imaging studies of membrane microstructure, lipid order, and membrane tension response in living cells[1][2][3].

(3S,4R)-ME-344 is the (3S,4R)-enantiomer of ME-344 (HY-112749). ME-344 is a mitochondrial Heme oxygenase 1 (HO-1) inhibitor. ME-344 specifically binds and alters HO-1 structure, and increases HO-1 translocation from the rough endoplasmic reticulum to mitochondria, but only in drug-sensitive cells (such as H460 and SHP-77 cells). ME-344 decreases mitochondrial ATP production and induces ROS, with subsequent disruption of redox homeostasis and mitochondrial function. ME-344 has significant antitumor activity, and can be used for cancers like breast cancer research .

Imiprothrin is an inducer that induces CYP1A2 and metallothionein 1a, with significant genotoxicity and cytotoxicity. In rat hepatocytes, Imiprothrin initiates detoxification responses by triggering the overexpression of these two genes. Imiprothrin induces chromosomal aberrations and micronucleus formation in rat bone marrow cells, and causes DNA damage in hepatocytes. Imiprothrin triggers oxidative stress in rats, leading to lipid peroxidation, excessive reactive oxygen species production and redox imbalance, which in turn impairs liver and kidney functions and causes tissue damage. Imiprothrin inhibits weight gain in mice, and even causes high mortality in female mice at high doses. However, it shows no carcinogenicity in rat experiments; among relevant indicators, aspartate aminotransferase and total protein are identified as sensitive toxicity biomarkers .

Potassium Channel

Research Areas:

Cardiovascular Disease

Aprikalim (RP 52891), a potassium channel opener (KCO), activates ATP-sensitive K+ (KATP) channels in guinea pig ventricular myocytes. Using patch-clamp techniques, it was found that aprikalim enhances KATP channel activity more effectively in Mg-NDP solution compared to standard solutions. In Mg-NDP solution, aprikalim reduced the sensitivity of KATP channels to ATP, increasing the concentration of ATP required to inhibit channel activity by half (K1) from 56 μM to 180 μM. However, this effect diminished over time. Aprikalim's ability to activate KATP channels in Mg-NDP solution suggests potential therapeutic implications in modulating cardiac excitability and may relate to changes in channel protein enzymatic activity under experimental conditions .
