Sulfo-ara-F-NMN purchased from MedChemExpress. Usage Cited in: Cell. 2025 Dec 11;188(25):7137-7154.e21.  [Abstract]

Response curves of intracellular cADPR production and NAD+ consumption in the HEK293T cells treated with Sulfo-ara-F-NMN (CZ-48) (1-4 mM). HEK293T cells stably expressing WT hSARM1 were treated with different doses of CZ-48 for 24 h. cADPR and NAD+ levels in the resultant cells were assessed by MS.

Sulfo-ara-F-NMN purchased from MedChemExpress. Usage Cited in: Cell. 2025 Dec 11;188(25):7137-7154.e21.  [Abstract]

Representative IF images of the noted mouse neural cells treated with dsDNA transfection or Sulfo-ara-F-NMN (CZ-48). The noted neural cells were treated with 2 μg/mL dsDNA transfection per 16 h or 1 mM CZ-48. After 60 h, the resultant cells were stained with anti-SARM1 antibody. CZ-48 promoted both WT and 3KE neural cell death, while dsDNA transfection promoted only WT cell death. BF, bright field; red, mSARM1; blue, DAPI. Scale bar, 10 μm.

Sulfo-ara-F-NMN purchased from MedChemExpress. Usage Cited in: Nat Chem Biol. 2025 Aug 22.  [Abstract]

    Sulfo-ara-F-NMN (CZ48) (1–1000 μM; 24 h) acted as an NMN-mimicking SARM1 activator and induced SARM1-dependent cell death in HEK-293T cells overexpressing SARM1dN.

Sulfo-ara-F-NMN purchased from MedChemExpress. Usage Cited in: iScience. 2024 May 8;27(6):109940.

MTT conversion to formazan at day 2 of PMA-differentiated THP-1 cells. Cells were treated with PMA for 24 h followed by being cultured in PMA-free, containing indicated stimulants media for 24 h. Unstimulated cells are used as normalization, set as 100%. DMSO is used as a control for Sulfo-ara-F-NMN (CZ-48) (100 μM; 24 h) and it’s also treated in cells shown as LPS. Sulfo-ara-F-NMN (CZ-48) treatment significantly reduced MTT conversion in SARM1-WT but not SARM1-E642A nor EV cells, both with or without LPS.

Left panel, PMA differentiated THP-1 cells were stimulated with LPS and/or Sulfo-ara-F-NMN (CZ-48) (100 μM) for 6 h. Immunoblots of pro-IL-1β and SARM1 (to show no effect by NADase in other proteins) from cell lysates of PMA-differentiated THP-1 stimulated with LPS and/or CZ-48. β-actin was used as a loading control. Right panel, quantification of pro-IL-1β.

    Description

Sulfo-ara-F-NMN (CZ-48) is a mimetic of nicotinamide mononucleotide (NMN). Sulfo-ara-F-NMN acts selectively, activating SARM1 but inhibiting CD38 (IC50 around 10 μM). Sulfo-ara-F-NMN induces intracellular cyclic ADP-ribose (cADPR) production[1].

    In Vitro

Sterile alpha and Toll/interleukin-1 receptor motif-containing 1 (SARM1) is an adaptor protein in the Toll-like receptor pathway. Sulfo-ara-F-NMN activates SARM1 to produce cyclic ADP-ribose and induces non-apoptotic cell death[1].

    In Vivo:

Select the appropriate dissolution method based on your experimental animal and administration route.

For the following dissolution methods, please ensure to first prepare a clear stock solution using an In Vitro approach and then sequentially add co-solvents:
To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for in vivo experiments, it is recommended to prepare freshly and use it on the same day.
The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.

    • Protocol 1

      Add each solvent one by one:  10% DMSO    40% PEG300    5% Tween-80    45% Saline

      Solubility: ≥ 2.5 mg/mL (7.10 mM); Clear solution

This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).

Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.

Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
    • Protocol 2

Add each solvent one by one:  10% DMSO    90% (20% SBE-β-CD in Saline)

      Solubility: ≥ 2.5 mg/mL (7.10 mM); Clear solution

This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).

Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.

Preparation of 20% SBE-β-CD in Saline (4°C, storage for one week): 2 g SBE-β-CD powder is dissolved in 10 mL Saline, completely dissolve until clear.

    Help & FAQs
    • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
