Description

W19-LNP is a lipid nanoparticle formulated with a Ugi-reaction-derived ionizable lipid (W19). W19-LNP demonstrates highly selective mRNA delivery to the spleen in female BALB/c mice upon intravenous administration. W19-LNP can be used for the research of mRNA-based vaccine delivery, immune cell reprogramming, and gene-engineered cell preparation[1].

In Vitro

W19-LNP (200 ng; 6 h) exhibit exceptional mRNA transfection efficiency in 16HBE and HepG2 cells[1].
W19-LNP (0.125-4.00 ng/µL; 24 h) show good in vitro safety in 16HBE cells, with relative cell viability remaining above 80%[1].

MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.

Cell Viability Assay[1]

Cell Line: 16HBE cells
Concentration: 0.125; 0.25; 0.50; 1.00; 2.00; 4.00 ng/µL
Incubation Time: 24 h
Result: Maintained cell viability above 80% at the highest tested concentration of 4.0 ng/µL.
In Vivo

W19-LNP (0.2 mg/kg; i.v.; single dose) demonstrate highly selective spleen-targeted mRNA delivery in healthy BALB/c mice[1].
W19-LNP (1 mg/kg; i.v.; single dose) exhibit good in vivo biocompatibility in healthy BALB/c mice, with no notable pathological tissue changes and biochemical parameters within normal safety ranges[1].

MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.

Animal Model: BALB/c (female, eight-week-old)[1]
Dosage: 0.2 mg/kg
Administration: i.v.; single dose
Result: Achieved high mRNA transfection potency.
Exhibited highly selective spleen targeting, with a proportion of splenic transfection (PST) of 95.4%.
Animal Model: BALB/c (female, six-week-old)[1]
Dosage: 1 mg/kg
Administration: i.v.; single dose
Result: Showed no remarkable pathological alterations in major tissues: uniform myocardial fibers with distinct boundaries in heart, indistinct liver lobule boundaries without inflammatory cell infiltration in liver, evenly distributed red pulp with clear white pulp boundaries in spleen, no significant vascular abnormalities in lung, and evenly distributed glomeruli without inflammatory cell infiltration in kidney.
Revealed statistically significant increases in aspartate aminotransferase (AST) and blood urea nitrogen (BUN) compared to PBS controls, but all values remained within the normal safety range.

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
