Guide (The following is a recommended experimental protocol for reference only and should be adjusted according to specific needs)
1. Solution preparation[3]
1.1 Stock solution storage
Storage: Aliquot and store protected from light at -20°C or -80°C. Avoid repeated freeze-thaw cycles.
1.2 Working solution preparation
Dilute with PBS or cell culture medium. The stock solution can be diluted as needed according to actual conditions. Note: if the solvent is DMSO, its cytotoxicity must be considered and a solvent control should be prepared; if the solvent is pure water, the working solution should be filtered and sterilized before being added to cells.
Note: The working solution should be freshly prepared, used immediately, and protected from light.
2. Cell staining
2.1 Seed cells into black 96-well microplates with clear bottoms.
2.2 Add Monochlorobimane (1-10 μM) and incubate for 30 min (optimize according to the experiment).
2.3 Measure fluorescence intensity using a fluorescence microscope or a fluorescence microplate reader (Ex/Em = 380/480 nm).
