Plusbacin A3
Plusbacin A3 is a cyclic lipodepsipeptide isolated from Pseudomonas sp. PB-6250 that inhibits bacterial cell wall biosynthesis and disrupts membrane integrity. Plusbacin A3 binds lipid intermediates, blocking transglycosylation and lipid intermediate formation in peptidoglycan synthesis. Plusbacin A3 inhibits teichoic acid biosynthesis. Plusbacin A3 exhibits activity against Gram-positive bacteria, including methicillin-resistant Staphylococcus aureus and vancomycin-resistant Enterococci, with low susceptibility to resistance, and is inactive against Gram-negative bacteria. Plusbacin A3 can be used in research on bacterial infections.

Activité biologique
Description
In Vitro
Plusbacin A3 exhibits antimicrobial activity with a MIC range of 0.2 to 6.25 μg/mL against S. pyogenes M49 strain NZ131, S. agalactiae strain A909, ATCC 33591, and ATCC 51299, but is inactive against Escherichia coli (ATCC 25922) and Pseudomonas aeruginosa (ATCC 27853)[2].
Plusbacin A3 (0.78-3.13 μg/mL) exhibits potent antibiotic activity against methicillin-resistant Staphylococcus aureus and VanA-type vancomycin-resistant enterococci with MIC values ranging from 0.78 to 3.13 μg/mL[3].
Plusbacin A3 inhibits N-acetylglucosamine incorporation into staphylococcal cell wall peptidoglycan with an IC50 close to its MIC value[3].
Plusbacin A3 inhibits both nascent peptidoglycan formation and lipid intermediate formation in bacterial cell wall biosynthesis at a site distinct from the Vancomycin (HY-B0671) binding domain[3].
Plusbacin A3 (20 h) exhibits strong antibacterial activity against methicillin-resistant Staphylococcus aureus and vancomycin-resistant enterococci, with MIC values ranging from 0.78 to 3.13 μg/mL[5].
Plusbacin A3 (30 min) inhibits the incorporation of [14C]GlcNAc into peptidoglycan of Staphylococcus aureus NCTC8325 whole cells with an IC50 of 0.62 μg/mL[5].
Plusbacin A3 (Various concentrations; 60 min) inhibits the formation of lipid intermediates (IC50 = 2.3 μg/mL) and nascent peptidoglycan (IC50 = 0.4 μg/mL) in an in vitro assay using Staphylococcus aureus SRM133 wall-membrane particulate and supernatant fractions[5].
The inhibition of transglycosylation by Plusbacin A3 (0.78 μg/mL; 60 min) in Staphylococcus aureus SRM133 fractions is not suppressed by acetyl-Lys-D-Ala-D-Ala[5].
The antibacterial activity of Plusbacin A3 (5 μg per disk; Overnight) against Staphylococcus aureus NCTC8325 is antagonized by a wall-membrane particulate[5].
